First report of Pestalotiopsis diospyri causing canker on persimmon trees Rev. Bras. Frutic.
Alves,Giselda; Verbiski,Francineia Silva; Michaelides,Themis J.; May-de Mio,Louise Larissa.
During 2006 to 2009 season symptoms of a canker disease were observed on twigs and branches of young and mature persimmon trees (Diospyros kaki L.) cv. Fuyu in the States of Santa Catarina and Paraná in the Southern Brazil. The cankers result in severe damage and reduced production. Isolations from the margins of these cankers revealed a genus of Pestalotiopsis. Koch's postulates were confirmed using two isolates of the pathogen which was identified as Pestalotiopsis diospyri.

Diurnal and seasonal variability in bird counts in a forest fragment in southeastern Brazil Zoologia
Antunes,Alexsander Z..
Diurnal variation in detection can influence bird census results. I measured variation in daily and seasonal detections of birds in a forest fragment at Barreiro Rico ranch, Anhembi, state of São Paulo. Birds were recorded on transects of unlimited distance, sampled one day per month, dawn to dusk, between December 2000 and March 2001 (rainy season) and between May and August 2001 (dry season). Significant daily variation in detection occurred during the rainy season, with most detections between dawn and 9 h. In the dry season detections declined for total numbers of individuals and species and most detections occurred during late morning and mid afternoon. Detection patterns might reflect seasonal variation in bird behavior rather than seasonal variation...

Development of genomic probes for detection of hepatitis A virus and enteroviruses in shellfish ArchiMer
Apaire, M; Leguyader, F; Besse, B; Billaudel, S.
The development of molecular biology techniques has modified research procedures for certain viruses, particularly hepatitis A virus (HAV) and enteroviruses. Accordingly, we designed cDNA and RNA genomic probes to detect viruses in shellfish in the environment. Better sensitivity was noted with RNA than cDNA probes: only 17/83 shellfish samples were positive with HAV and enterovirus DNA probes, where as 42/83 were positive with HAV and enterovirus RNA probes.

A long term study of bonamiosis in Quiberon bay, France ArchiMer
Arzul, Isabelle; Miossec, Laurence; Blanchet, Estelle; Garcia, Celine; Joly, Jean-pierre; Francois, Cyrille; Berthe, Franck.
Bonamiosis was first reported in association with mass mortality of flat oysters, Ostrea edulis, in June 1979 in L'Ile Tudy, Brittany. The disease rapidly spread to all of the oyster farming areas in France but also in other European countries. The French flat oyster production, which already suffered from another protozoan disease, marteiliosis, decreased from 5500 mt in 1979 to less than 2000 mt after 1980. In 2001, about 350 French farms sell 1650 mt of flat oysters. This production mainly relies on natural spat collection which specially occurs in Quiberon bay. One third of this spat is transferred from South to North Brittany for further growth. Quiberon bay constitutes an interesting site regarding the surveillance of bonamiosis because of the...

Quick adaptation of Ralstonia Solanacearum to copper stress to recover culturability and growth in water and soil BJM
Ascarrunz,Sergio Daniel Moreira; Natsuaki,Tomohide; Honjo,Hitoshi; Fukui,Ryo.
Cells of Ralstonia solanacearum were exposed to Cu in distilled water, and the resulting Cu-stressed non-culturable cells were inoculated to natural (non-pasteurized) and pasteurized soils in order to examine their culturability and recovery. Exposing the cells to 20 µM CuSO4 produced transitory non-culturable cells, which exhibited a remarkable recovery in culturability after incubation in the solution for 36 h, reaching a density near the initial level by 108 h. To determine whether such non-culturable cells actually "resuscitated" or multiplied after adapting to Cu toxicity, growth curves were constructed in order to contrast the rates of increase in culturable cell numbers between Cu-stressed or non-stressed inocula. Additionally, fresh non-stressed...

Molecular detection of Potato yellow vein virus in the natural whitefly vector Trialeurodes vaporariorum, Westwood Rev. Protección Veg.
Barragan,Carlos Eduardo; Guzmán-Barney,Mónica.
Potato yellow vein virus (PYVV) expresses yellowing symptoms on plants and it is transmitted by the whitefly vector Trialeurodes vaporariorum Westwood. No molecular viral detection from the vector extracts has been reported to date. The objectives of this study were I) amplification of the PYVV major coat protein (CP) gene from viruliferous vector extracts by RT-PCR using specific primers, II) cloning and III) sequencing and sequences analysis. Some Colombian Solanum phureja (Jus et Buk) field potato plants expressing yellowing symptoms and confirmed PYVV positive by RT-PCR were used as virus donor plants for feeding whiteflies. Viruliferous vectors were obtained by depositing aviruliferous whiteflies on symptomatic leaves and kept in a muslin...

Detection of ostreid herpesvirus 1 DNA by PCR in bivalve molluscs: A critical review ArchiMer
Batista, Frederico; Arzul, Isabelle; Pepin, Jean-francois; Ruano, Francisco; Friedman, Carolyn S.; Boudry, Pierre; Renault, Tristan.
Herpes-like viral infections have been reported in different bivalve mollusc species throughout the world. High mortalities among hatchery-reared larvae and juveniles of different bivalve species have been associated often with such infections. The diagnosis of herpes-like viruses in bivalve molluscs has been performed traditionally by light and transmission electron microscopy. The genome sequencing of one of these viruses, oyster herpesvirus 1 (OsHV-1), allowed the development of DNA-based diagnostic techniques. The polymerase chain reaction (PCR) has been used for the detection of OsHV-1 DNA in bivalve molluscs at different development stages. In addition, the PCR used for detection of OsHV-1 has also allowed the amplification of DNA from an OsHV-1...

Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer
Batista, Frederico; Taris, Nicolas; Boudry, Pierre; Renault, Tristan.
A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.

Evaluation of the Direct and Enrichment Iso 10272 Methods for the Detection of Campylobacter Spp. in Chicken Meat Rev. Bras. Ciênc. Avic.
Benetti,TM; Abrahão,WM; Ferro,ID; Macedo,REF; Oliveira,TCRM.
ABSTRACT In 2006, the International Organization for Standardization (ISO) published two standard protocols for the detection and enumeration of Campylobacter spp. in foods: the ISO 10272-2(direct method) and the ISO 10272-1 (enrichment method). The aim of this study was to compare the efficiency of these two methods in the detection of Campylobacter spp. and propose a modification in the enrichment method. Compared with the enrichment method, the direct method yielded a higher number of positive results for Campylobacter spp. and, consequently, presented higher sensitivity percentage. The isolation of Campylobacter spp. was more difficult when 10 mL of rinse was used in the enrichment method, which is currently recommended by the ISO protocol. Therefore,...

Foodborne viruses: Detection, risk assessment, and control options in food processing ArchiMer
Bosch, Albert; Gkogka, Elissavet; Le Guyader, Soizick; Loisy-hamon, Fabienne; Lee, Alvin; Van Lieshout, Lilou; Marthi, Balkumar; Myrmel, Mette; Sansom, Annette; Schultz, Anna Charlotte; Winkler, Anett; Zuber, Sophie; Phister, Trevor.
In a recent report by risk assessment experts on the identification of food safety priorities using the Delphi technique, foodborne viruses were recognized among the top rated food safety priorities and have become a greater concern to the food industry over the past few years. Food safety experts agreed that control measures for viruses throughout the food chain are required. However, much still needs to be understood with regard to the effectiveness of these controls and how to properly validate their performance, whether it is personal hygiene of food handlers or the effects of processing of at risk foods or the interpretation and action required on positive virus test result. This manuscript provides a description of foodborne viruses and their...

Potential ways to address the issues that relate to the techniques for analyzing food and environmental samples for the presence of enteric viruses are discussed. It is not the authors' remit to produce or recommend standard or reference methods but to address specific issues in the analytical procedures. Foods of primary importance are bivalve molluscs, particularly, oysters, clams, and mussels; salad crops such as lettuce, green onions and other greens; and soft fruits such as raspberries and strawberries. All types of water, not only drinking water but also recreational water (fresh, marine, and swimming pool), river water (irrigation water), raw and treated sewage are potential vehicles for virus transmission. Well over 100 different enteric viruses...

Observing and Studying Extreme Low Pressure Events with Altimetry ArchiMer
Carrere, Loren; Mertz, Francoise; Dorandeu, Joel; Quilfen, Yves; Patoux, Jerome.
The ability of altimetry to detect extreme low pressure events and the relationship between sea level pressure and sea level anomalies during extra-tropical depressions have been investigated. Specific altimeter treatments have been developed for tropical cyclones and applied to obtain a relevant along-track sea surface height (SSH) signal: the case of tropical cyclone Isabel is presented here. The S- and C-band measurements are used because they are less impacted by rain than the Ku-band, and new sea state bias (SSB) and wet troposphere corrections are proposed. More accurate strong altimeter wind speeds are computed thanks to the Young algorithm. Ocean signals not related to atmospheric pressure can be removed with accuracy, even within a Near Real Time...

Use of cardiopulmonary exercise testing to assess early ventilatory changes related to occupational particulate matter BJMBR
Chao,T.P.; Sperandio,E.F.; Ostolin,T.L.V.P.; Almeida,V.R.; Romiti,M.; Gagliardi,A.R.T.; Arantes,R.L.; Dourado,V.Z..
Spirometry has been used as the main strategy for assessing ventilatory changes related to occupational exposure to particulate matter (OEPM). However, in some cases, as one of its limitations, it may not be sensitive enough to show abnormalities before extensive damage, as seen in restrictive lung diseases. Therefore, we hypothesized that cardiopulmonary exercise testing (CPET) may be better than spirometry to detect early ventilatory impairment caused by OEPM. We selected 135 male workers with at least one year of exposure. After collection of self-reported socioeconomic status, educational level, and cardiovascular risk data, participants underwent spirometry, CPET, body composition assessment (bioelectrical impedance), and triaxial accelerometry (for...

ABSTRACT Many countries in the Americas have detected local transmission of multiple arboviruses that cause febrile illnesses. Therefore, laboratory testing has become an important tool for confirming the etiology of these diseases. The present study aimed to compare the sensitivity and specificity of three different Zika virus detection assays. One hundred serum samples from patients presenting with acute febrile symptoms were tested using a previously reported TaqMan® RT-qPCR assay. We used a SYBR® Green RT-qPCR and a conventional PCR methodologies to compare the results. Of the samples that were determined to be negative by the TaqMan® RT-qPCR assay, 100% (Kappa = 0.670) were also found to be negative by SYBR® Green RT-qPCR based on Tm comparison;...

Detection de fissure sur structure en mer en presence de salissure marine projet "Crabe" ArchiMer
Daste, S.
The "CRABE" method is a crack detection technique based on the use of low electromagnetic frequencies. This (patented) method relies on a global injection of current within the structure under inspection. The presence of a crack shall cause a disruption in the current flow, and consequently of the magnetic field off the surface in the vicinity of a defect. It is this magnetic signature which is detected by the magnetic sensor used with "CRABE". The major advantage of this process, developed by TRAVOCEAN and the LETI Division of CEA with the help of IFREMER, appears clearly: No preliminary cleaning of the welds to be inspected is required, since no contact is necessary. Similarly, it shall be possible to inspect the whole of structures to a total cost all...

Garlic viral complex: identification of Potyviruses and Carlavirus in Central Brazil Trop. Plant Pathol.
FAJARDO,THOR V. M.; NISHIJIMA,MARTA; BUSO,JOSÉ A.; TORRES,ANTÔNIO C.; ÁVILA,ANTÔNIO C.; RESENDE,RENATO O..
Garlic viruses often occur in complex infections in nature. In this study, a garlic virus complex, collected in fields in Brazil, was purified. RT-PCR was performed using specific primers designed from the consensus regions of the coat protein genes of Onion yellow dwarf virus, a garlic strain (OYDV-G) and Leek yellow stripe virus (LYSV). cDNA of Garlic common latent virus (GCLV) was synthesized using oligo-dT and random primers. By these procedures individual garlic virus genomes were isolated and sequenced. The nucleotide sequence analysis associated with serological data reveals the presence of two Potyvirus OYDV-G and LYSV, and GCLV, a Carlavirus, simultaneously infecting garlic plants. Deduced amino acid sequences of the Brazilian isolates were...

Rapid detection and quantification of the marine toxic algae, Alexandrium minutum, using a super-paramagnetic immunochromatographic strip test ArchiMer
Gas, Fabienne; Baus, Beatrice; Quere, Julien; Chapelle, Annie; Dreanno, Catherine.
The dinoflagellates of Alexandrium genus are known to be producers of paralytic shellfish toxins that regularly impact the shellfish aquaculture industry and fisheries. Accurate detection of Alexandrium including A. minutum is crucial for environmental monitoring and sanitary issues. In this study, we firstly developed a quantitative lateral flow immunoassay (LFIA) using super-paramagnetic nanobeads for A. minutum whole cells. This dipstick assay relies on two distinct monoclonal antibodies used in a sandwich format and directed against surface antigens of this organism. No sample preparation is required. Either frozen or live cells can be detected and quantified. The specificity and sensitivity are assessed by using phytoplankton culture and field samples...
